In your previous questin, I referred to the Jewish rebellion that was put down by Vespasian, who would later become an emperor of Rome.  While he was putting this rebellion down, he also got most of the workers who would build the Colosseum.  The video says (also right around the 1 minute mark) that it was slaves taken by Vespasian who did the work of building the Colosseum.

The video says that about 12,000 slaves did most of the work.  It says that they were pretty much worked to death in building the arena.  For example, they had to drag huge blocks of rock from a quarry 20 miles away.
