The effective method that helps to isolate the microbial colonies is called streak plate method. This method helps to isolate and sample the individual bacterial colonies by growing bacteria on a growth media surface.

The streaking procedure uses pure microbial cultures and the transfer of isolated colonies from a gelating plate to other gelatin plate is done using a sterile swab. The sterilization of swab is done using a Bunsen burner and it is performed whenever different zones are streaked. The result of iterated sterilizations between streaked regions is decrease in number of microorganisms that are deposited in the streaking process.

There exists two method used in the streak plate procedure called four quadrant streak and three sector - T streak.
