That ladder is composed of a "backbone," forming the sides of the ladder and consisting of alternating sugar and phosphate groups, and a bonded base pair. These bases pair in predictable and specific ways to form the rungs of that ladder.

The four bases in DNA are adenine (A), cytosine (C), guanine (G), and thymine (T). Using hydrogen bonds, each base will pair with a specific corresponding base.

Adenine and thymine will form a bond.

Cytosine and guanine will form a bond.

So, by being given one half of a DNA sequence, you can predict the corresponding bases because of this predictable pairing. If you have an A, it will match with a T. If you have a T, it will match with an A. If you have a C, it will match with a G. If you have a G, it will match with a C.
