Southern blot analysis is used for the detection of DNA or gene sequence in circular or large DNA. This can also be used to measure the molecular weight and compare relative amounts of different samples.


Where as RFLP (Restriction Fragment Length Ploymorphism) is used for compare the DNA fragments after reaction with restriction enzymes. This is used to detect gentic mutations and diseases where mutations have destroyed restriction sites.
