While not "doomed" in a strict sense, the odds are certainly stacked against the pair. The trouble they have with communication would have certainly led to trouble without the intervention of Iago, but it would be difficult to say it would have doomed them.
When a couple has the sort of dynamic conflict that Desdemona and Othello do (difference in age, race, social class, father-daughter dynamic, social training, differing levels of naivete and trust), maintaining a relationship becomes an uphill battle. Othello and Desdemona's relationship was unstable to begin with, but "doomed" is probably stretching it.
It is important to note how easy it was for Iago to manipulate the situation -- that speaks volumes about the strength of the relationship and how secure Othello and Desdemona feel.
