Iago tells Cassio that "the General's wife is now the general," meaning that she is now in charge of him and thus in charge of the decisions he makes concerning the State.  Furthermore, he comments on the fact that Othello is a trusting man and believes in the good of mankind.  He also feels that Othello will do whatever it takes to make her happy, and these two facts coupled together make him an easy target for manipulation.
