In embryonic stem cells technique, embryo derived stem cells are cultured. These stem cells are transfected with a vector containing the desirable gene. This allows targeting of gene to a particular site by homologous recombination.
Somatic cells are good alternative. Somatic embryogenesis in culture was demonstrated for the first time in carrots, where bipolar embryos developed from single cells. Further, the tissue derived from these somatic embryos spontaneously developed into embryos again.
Precursor or stem cells, which are capable of proliferation, but remain undifferentiated until the correct inducing conditions are applied so that some or all of the cells mature to differentiated cells, these are pluripotent cells.
