The refrain from Eminem's "Lose Yourself" defines Macbeth's moment where he has to make a life-altering decision. After being told my the Weird Sister that he will become king, he must make this choice of whether or not to murder Duncan. In an attempt to fulfill her own selfish and greedy ambitions, Lady Macbeth encourages her husband to commit the deed and allow the night guards to take the blame for the murder.

Macbeth reluctantly agrees to murder his king and knows this opportunity only comes once. He must kill Duncan while Duncan is staying as a guest in his home because Macbeth will never get another chance to become king.
